Determination of tetracycline residues in chicken by polymer monolith microextraction and sweeping micellar electrokinetic chromatography with cation-selective exhaustive injection

摘要:

以聚(甲基丙烯酸-乙二醇二甲基丙烯酸酯)毛细管整体柱为萃取介质,考察了其对强力霉素和金霉素的萃取性能,将其与阳离子选择性耗尽进样-胶束扫集在线富集技术联用,建立了一种测定鸡肉中强力霉素和金霉素两种四环素类药物残留的分析方法。方法检出限和定量限分别为3.4 ng/g,12.6ng/g和11.4 ng/g,42.0 ng/g。在50~1000 ng /g的范围内峰面积与样品浓度具有良好的线性关系,日内、日间测定的相对标准偏差不高于8.2%,加标回收率在86.9%~91.3%之间。该方法操作简便、灵敏度高且成本低,重现性好,可用于鸡肉中痕量强力霉素和金霉素残留的测定。

Abstract:

A poly (methacrylic acid-ethylene glycol dimethacrylate) monolithic capillary column was prepared and applied as an extraction device, the extraction performance of doxycycline and chlortetracycline was investigated. By combining the polymer monolith mcroextraction (PMME) and sweeping micellar electrokinetic chromatography with cation-selective exhaustive injection(CSEI),the method for the quantitative monitoring of doxycycline and chlortetracycline residues in chicken was established. To obtain the optimum extraction efficiency, several parameters relating to MEKC (such as pH, buffer concentration, SDS concentration, separation voltage), CSEI (sample solvent, injection voltage and time, water plug length)and PMME (including extraction volume, pH value of the sample matrix and sample solvent) were systematically investigated. Under the optimum conditions, low detection limits (3.4 ng/g for doxycycline and 12.6 ng/g for chlortetracycline) and quantification limits (11.39ng/g for doxycyline and 41.99ng/g for chlortetracycline) were obtained. The proposed method showed good linearity ranging from 50 to 1000 ng/g. The recoveries for fortified chicken samples were ranged from 85.1 to 91.7%. Excellent reproducibility of the method was exhibited by intra- and inter-day precisions with the relative standard deviations less than 8.2%. The proposed method was simple, sensitive, accurate and low cost, and it can be applied to simultaneously determine the doxycycline and chlortetracycline residues in chicken samples.
