槲寄生中高圣草素-7-O-β-D-葡萄糖苷的分离及含量测定

Isolation and Determination of Homoeriodictyol-7-O-β-D-Glycoside in Viscum coloratum

摘要: 对槲寄生中的有效成分高圣草素-7-O-β-D-葡萄糖苷进行了分离和结构鉴定,并对其含量进行了分析。色谱条件:C18柱(200 mm×4.6 mm i.d., 5 μm),流动相为乙腈-0.5%冰醋酸水溶液(体积比为18∶82),流速为1.0 mL/min,柱温为30 ℃,检测波长为284 nm,进样量为10 μL。结果表明,高圣草素-7-O-β-D-葡萄糖苷的峰面积与其质量浓度有良好的线性关系,相关系数r为0.9997;方法的加标回收率为96.0%~100.1%。该方法简便、快速、准确,精密度好,可作为槲寄生质量控制的一个有效方法。

Abstract: The homoeriodictyol-7-O-β-D-glycoside was isolated from Viscum coloratum and identified by mass spectrometry and nuclear magnetic resonance (NMR) (1H NMR and 13C NMR). A method for determination of homoeriodictyol-7-O-β-D-glycoside in Viscum coloratum was developed by using a Kromasil C18 column (200 mm×4.6 mm i.d., 5 μm) with a mixture of acetonitrile and 0.5% glacial acetic acid solution (18∶82, v/v) as mobile phase at a flow rate of 1.0 mL/min. The detection wavelength was set at 284 nm and temperature was set at 30 ℃. The volume of injection was 10 μL. Good linear relationship (r=0.9997) between the mass concentration and the peak area of homoeriodictyol-7-O-β-D-glycoside was obtained in the range of 1.0-32.0 mg/L. The recoveries were found to be in the range of 96.0%-100.1%. The results of the experiments demonstrated that the established method is rapid and simple with good accuracy and reproducibility. The method is suitable for the quality control of Viscum coloratum from different sources.
