Abstract:
采用高效液相色谱法(HPLC)同时测定了豆制品中的碱性橙和碱性嫩黄O染料的含量
,样品用乙腈提取后用正己烷脱脂净化,然后在碱性条件下用乙醚萃取,最后溶于甲醇中进
行HPLC分离、检测。所使用的色谱柱为Gemini C18(5 μm,4.6 mm×150 mm),紫外检
测波长分别为458 nm(碱性橙)和436 nm(碱性嫩黄O),流动相为0.03%磷酸溶液/甲醇
,梯度洗脱。碱性橙的检出限为0.01 mg/kg,碱性嫩黄O的检出限为0.02 mg/kg。回收率
为71%~80%,相对标准偏差(RSD)为2.2%~5.0%(n=6),两种染料在0.2~4.0 μg/mL内呈
良好的线性关系。
