Enantiomeric separation of liarozole on amylose chiral stationary phase

Abstract: A high performance liquid chromatographic method was established for the enantiomeric separation of liarozole. Baseline chiral separation of liarozole was achieved under normal-phase chromatographic mode by the Chiralpak AD-H chiral stationary phase. The influences of the concentration and kind of organic solvent, the proportion of acid and base, column temperature and flow rate on the enantiomeric separation were studied. The optimized chromatographic conditions were hexane-ethanol (containing 0.3% diethylamine and 0.1% glacial acetic acid) (80∶20, v/v) as the mobile phase with a flow rate of 0.6 mL/min and detection at the wavelength of 254 nm. The column temperature was set at 20 ℃. The resolution of 3.4 for liarozole was achieved under the above chromatographic conditions. The method is simple, rapid and with good reproducibility.
